T1 - Liposome-embedded heme - totally synthetic oxygen carrier and its oxygen carrying capacity

N2 - Synthetic heme was embedded in bilayers of liposome to produce a totally artificial oxygen-carrying substance. The in vivo oxygen-carrying capacity of this liposome-embedded heme (L/H) was studied in dogs. Dogs weighing 8 to 10 kg were anesthetized, intubated and mechanically ventilated with ambient air. A Swan-Ganz catheter and an arterial line were inserted for blood sampling and pressure monitoring. The hemoglobin (Hb) and L/H concentration in the blood were measured. After injection of L/H, cardiac output, Hb and L/H concentration in the blood were measured and oxygen transport by Hb and L/H, oxygen consumption from Hb and L/H, oxygen volume transported in the total oxygen transport, and oxygen volume consumed from L/H in the total oxygen consumption were calculated. It was found that L/H transported oxygen to the tissue, and that oxygen volume transported by L/H and oxygen volume consumed from L/H were proportional to L/H.
