Prospects for the structural analysis of biological specimens by X-ray diffraction imaging

As described in Chap. 9, X-ray diffraction imaging (XDI) has the potential to reconstruct the three-dimensional (3D) electron density maps of biological specimens. The recent XDI structural analyses allow us the opportunity to discuss future prospects for the 3D structural analyses of biological specimens in XDI experiments using either synchrotron X-rays or X-ray free electron laser (XFEL) pulses. Here, we consider the achievable resolution, taking into account the radiation damage in synchrotron XDI experiments and the heterogeneity of specimens in XFEL-XDI experiments.
