Three dimensional closed microchamber for evaluation of shapes effect on stem cell differentiation

We report polyethylene glycol diacrylate (PEGDA) three-dimensional (3D) closed microchambers for assaying 3D-shape-driven differentiation induction of stem cells. We succeeded in fabricating PEGDA microchambers of various 3D shapes. We then cultured mouse neural stem cells (mNSCs) in the microchambers that was closed with by an agarose cover. We confirmed the high sealability of the closed microchambers and that the shapes of cultured mNSCs followed the 3D shape of the microchambers. We believe that our PEGDA microchambers would control 3D shapes of tissues and be useful for examining 3D-shape-driven differentiation induction of stem cells.
